Default PCIex1 question

I want to put a tv tuner in my puter. It's pci2x1 interface. That wouldn't matter that my pciex16 card is being used is it? I just wanted to make sure. I thought maybe that you only can use one pcie slot or the other; not both.

Yeah you can use both slots.
